SPOILER ALERT for those who haven't seen the filtered video that got leaked a couple days ago.
Characters
Sam (Norman Reedus): He is the protagonist in the story, he seems to have a very strong bound with both Bridget and her cause of connecting all cities of the U.S again. He has been working as a delivery man (whatever that means in Death Stranding) for the U.S in the past. Fragile implies in one of the videos that Sam has DOOM´s, which seems to be some kind of mutations acquired by some people when they died and come back to life. For some reason after Bridget dies, he lost interest in the U.S government and gave up his work. He seems to be in a middle ground when it comes to sympathizing with Amelie and Die Hard Man.
He uses a B.B to sense B.T´s.
B.B (Bridge Baby): These babies help humans to create a connection with "the other side" and so he can feel the B.T's. Sam is able to see the B.T's when he is connected to the baby because the baby is "connected" to the womb of a clinically dead mother which body is maintained and therefore the baby has a connection with both the living and the B.T´s world. Sam has to connect the container where the baby lives from time to time to a machine that simulates this kind of wombs to fulfill its function in the best possible way.
B.T's: They seem to be entities that are neither alive or dead, they seem to have a connection with the world of the living (they can interact with the living). It seems that B.T can be connected through an “umbilical cord” to either, the black figure that appears in the trailers or to living people like Mama.
They can detect humans by sound and breathing. Humans prefer to commit suicide then to be caught by "dark" B.T.s, which implies that these kind of B.T´s are not dead but controlled by others and in a state of suffering.
The BT's can be connected or stranded and those that are connected can be connected to the world of the living (such as the BT that lives with Mama and that is transparent in color) or connected to the dark giant entity with cords that come from his fingers (making B.T´s dark and harmful).
Bridget: It's the name of the president of the United States, mother of Amelie. She died and now Amelie wants to continue her legacy of connecting all the cities of the United States.
Cliff (Mads Mikkelsen): Cliff is part of the "Homo Demens" and is portrayed as the antagonist in Death Stranding. He seems to have the power of connecting to stranded "souls" and control them in any way. From the trailers as the rest of Homo Demens he seems to have a connection or a deal with the "black entities" which allows him to use some powers. In one trailer he takes a cigarette and to light it he just raises his hand and it lights up like if an occult being was taking care of him.
Amelie (Lindsay Wagner): Daughter of the president of the United States (now dead) and the one chosen to continue her mother's mission to connect all the cities of the United States and thus be able to create communication between all the inhabitants. In the past, Amelie has already started a mission in which she traveled west to reconnect cities and people, but she was captured and the team she was going with was razed.
She is not only trying to complete her mother´s mission but also become the new president of the U.S "Samantha America Strand". (kevsmith202)
Die Hard Man (Tommie Earl Jenkins ): He seems to be a supporter of the cause of the former president and now of Amelie and a faithful helper for this cause.
Dead Man (Guillermo del Toro): Dead man remains a very mysterious character, the only thing we know so far is that he and Sam get along and all points in the direction that during the story he will help Sam and the U.C.A since he is part of Bridges.
Mama (Margaret Qualley): We know very little about her, only thing we know is that she is part of BRIDGES and takes care of what she calls her daughter which is a B.T. This B.T is linked to her which makes it not dangerous since it is not a stranded (disconnected) B.T which could lead in to it becoming a dark entity (dangerous).
She is a mechanic for BRIDGES (RamonesRazor).
Heartman (Nicolas Winding): As Deadman and Mama this is a character that remains a mystery for the lack of information. Like them he seems to help BRIDGES and Sam. He is a particular character because he seems to be dead but is able to come back to life for periods of 21 minutes every now and then. He says he is looking for his family in the beach. From his own words in his character trailer presentation “My body is present but my soul is in the beach”.
Fragile (Léa Seydoux): Fragile is a character that similar to the rest of them we have very few info about. From the trailer we can get that she seems to be sympathetic with Sam. She carries an umbrella probably to avoid time fall and eats Cryptobites to "keep the time fall away". She seems to see something of value in Sam since she ask him to work with her. In one trailer she says that like Sam she has Doom´s (Death Obtained Organic Mutations). (Rydael)
Organizations / Communities
U.C.A: It is the name by which Amelie supporters refer to the new government they want to create in the United States (United Cities of America).
Bridges: It is a group of people (Sam, Dead Man, Mama and Heartman for now) who seem to be independent but working for the U.S government for now. Their main objective is to connect the different cities of the U.S and as their logo says create a "Geo Surveillance System" (kevsmith202).
Homo Demens: They are the dark characters who also carry B.B´s (such as Cliff "Mad Mikkelsen") and who form an independence group who do not want to have anything to do with the UCA or with their plans, they have abducted Amelie in Edge Knot City. Die Hard Man defines them as a group of terrorists who go around the cities killing people and leaving craters. They seem to have some friendship or control over the B.T.s. It is said that they do not have any organization among them only a common ideology which is not named.
They seem to have a connection with the other side or kind of a "deal" with the "dark entities" that gives them special abilities like flying or controlling stranded souls for their own purposes.
Locations
Edge Knot City: It seems to be a place of refuge and the symbolic capital for a group of people who do not want to have anything to do with any type of government and are only governed by their own standards. They have Amelie locked up in this city to ensure this independence.
Knot city seems to be divided into different zones since "Knot Central" is named in a video and they refer to this area as a destroyed one and different from Edge Knot City. Also Capital Knot City is named in the Dead Man video, probably different areas of the same city.
Others
Sam's mission (for now): Travel west to reconnect the United States and rescue Amelie from Edge Knot City.
Time Fall: This kind of rain is one of the most characteristic things in the game. This rain has the particularity of speeding up the time of what ever it touches from human skin, plants to what ever has an organic nature. Form what we have seen in the videos Time Fall is an indicative of "dark" B.T´s being in the area. Fragile says that she eats Cryptobites to keep the time fall away, we will have to wait a bit more to give meaning to that sentence.
Craters: Craters are a significant part of the history of Death Stranding, these seem to be created thanks to a technology capable of manipulating the vacuum (vacuum engineering) This technology seems to be the substitute for nuclear bombs since it is talked about in the same terms as today we talk about nuclear bombs in that they are a dangerous weapon because of the immediate and prolonged consequences of using them.
Q-Pid: It is the pendant with mathematical formulas that Sam is wearing around his neck which is given by Die Hard Man since it is necessary to activate the terminals in the different cities that Sam has to visit to connect them again together. Probably the Q-Pid pronunciation sounds intentionally like Cupid since both of them are meant to bring people together.
The beach: Being on the beach or being stranded seems to be the name given to the state of being in limbo or being disconnected from both the world of the dead and the living. What implies that the ocean in Death Stranding is a metaphor for connection and union between people.
I came to this conclusion because Amelie says in a video not to grow old because her body is on the beach and in Mama's video she tells Sam not to fear the BT that is in that room because he is connected to her and not “Stranded” like the rest.
